    Aspects of functional variations of domination in graphs.

A signed k-subdominating function (signed kSF) of G is defined as a function f : V > {-I, I} such that f(N[v]) > 1 for at least k vertices of G, where N[v] denotes the closed neighborhood of v. The signed k-subdomination number of a graph G, denoted by yks-11(G), is equal to min{f(V) I f is a signed kSF of G}. If instead of the range {-I, I}, we require the range {-I, 0, I}, then we obtain the concept of a minus k-subdominating function. Its associated parameter, called the minus k-subdomination number of G, is denoted by ytks-101(G). In chapter 2 we survey recent results on signed and minus k-subdomination in graphs. In Chapter 3, we compute the signed and minus k-subdomination numbers for certain complete multipartite graphs and their complements, generalizing results due to Holm [30]. In Chapter 4, we give a lower bound on the total signed k-subdomination number in terms of the minimum degree, maximum degree and the order of the graph. A lower bound in terms of the degree sequence is also given. We then compute the total signed k-subdomination number of a cycle, and present a characterization of graphs G with equal total signed k-subdomination and total signed l-subdomination numbers. Finally, we establish a sharp upper bound on the total signed k-subdomination number of a tree in terms of its order n and k where 1 < k < n, and characterize trees attaining these bounds for certain values of k. For this purpose, we first establish the total signed k-subdomination number of simple structures, including paths and spiders. In Chapter 5, we show that the decision problem corresponding to the computation of the total minus domination number of a graph is NP-complete, even when restricted to bipartite graphs or chordal graphs. For a fixed k, we show that the decision problem corresponding to determining whether a graph has a total minus domination function of weight at most k may be NP-complete, even when restricted to bipartite or chordal graphs. Also in Chapter 5, linear time algorithms for computing Ytns-11(T) and Ytns-101(T) for an arbitrary tree T are presented, where n = n(T). In Chapter 6, we present cubic time algorithms to compute Ytks-11(T) and Ytks-101l(T) for a tree T. We show that the decision problem corresponding to the computation of Ytks-11(G) is NP-complete, and that the decision problem corresponding to the computation of Ytks-101 (T) is NP-complete, even for bipartite graphs. In addition, we present cubic time algorithms to computeYks-11(T) and Yks-101(T) for a tree T, solving problems appearing in [25]

    Dairy cows value an open area for lying down

    As dairy cows are being housed for longer periods, with all-year-round housing growing in popularity, it is important to ensure housed environments are meeting the needs of cows. Dairy cows are motivated to access open lying areas, although previous motivation studies on this topic have confounded surface type and location (i.e. pasture outdoors vs cubicles indoors). This study measured cow motivation for lying down on an indoor open mattress (MAT; 9 m x 5 m) compared to indoor mattress-bedded cubicles, thus removing the confounding factor of surface type and location. This was repeated for an identically sized indoor deep-bedded straw yard (ST), to investigate whether surface type affected motivation for an open lying area. Thirty Holstein-Friesian dairy cows were housed in groups of 5 (n = 5 x 6) in an indoor robotic milking unit with access to six mattress-bedded cubicles. To assess motivation, cows were required to walk increasing distances via a one-way indoor raceway to access the open lying areas: Short (34.5 m), followed by Medium (80.5 m) and Long (126.5 m). Cows could choose to walk the raceway, leading to the MAT or ST, to lie down or they could lie down on the cubicles for ‘free’. Overall, cows lay down for longer on the open lying areas at each distance compared to the cubicles, with cows lying down slightly longer on ST than MAT, although lying times on the open lying areas did decrease at the Long distance. However, cows were still lying for >60% of their lying time on the open lying areas at the Long distance. This study demonstrates that cows had a high motivation for an open lying area, the provision of which could better cater for the behavioural needs of housed dairy cows and improve housed dairy cow welfare

    Open-ended project work : sharpening students skills

    WMG delivered and supported a group project for the Experience Warwick Summer School run by the Warwick Outreach and Widening Participation team. Each group was given the same set of equipment and minimal guidance towards what to create: they were provided with challenge cards to spark imagination but, after the first day, these cards were hardly used with students instead chasing their own inspiration. Participants were supported by University of Warwick student ambassadors and mentors from the WMG Graduate Programme and WMG research staff during the project. Ambassadors coached the students on team work while the WMG mentor helped with technical aspects and realising their imaginative ideas. A showcase, attended by teachers and families, included a smart city model, a radio-based game and several remote-controlled or line-following vehicles. Two main outcomes from student self-reflections were discernible: 1. The students self-reported an increase in engineering-related skills. 2. Students became more aware of current engineering research areas and the role that research has in shaping modern society. This was a successful pilot of a project-based programme of activities for year 10 students. In the summer of 2020 this project will be repeated for a new cohort of year 10 students but also expanded into a full, engineering-based work experience programme

    Developing a patient-centred tool for pain measurement and evaluation in autosomal dominant polycystic kidney disease.

    BACKGROUND: Pain affects 60% of the autosomal dominant polycystic kidney disease (ADPKD) population. Despite being an early and debilitating symptom, it is poorly characterized and management is suboptimal. This study aimed to develop an ADPKD-specific pain assessment tool (APAT) to facilitate pain research. METHODS: Following a systematic review of PATs used in ADPKD studies and against international recommendations for pain trials, our multi-disciplinary team of clinical experts and patients constructed an ADPKD-pain conceptual framework of key pain evaluation themes. We compiled a new APAT covering domains prioritized within our framework using components of questionnaires validated in other chronic pain disorders. The APAT was administered longitudinally within a randomized high-water intake trial (NCT02933268) to ascertain feasibility and provide pilot data on ADPKD pain. RESULTS: Thirty-nine ADPKD participants with chronic kidney disease Stages 1-4 provided 129 APAT responses. Each participant completed a median of 3 (range 1-10) assessments. Respondents' mean ± standard deviation age was 47 ± 13 years; 59% (23) were female; and 69% (27) had enlarged kidneys with median time from diagnosis 14.2 (interquartile range 7.0-25.9) years. Pain (52%) and associated analgesic use (29%) were common. Pain severity was associated with increasing age [odds ratio (OR) = 1.07, P = 0.009], female gender (OR = 4.34, P = 0.018), estimated glomerular filtration rate <60 mL/min/1.73 m2 (OR = 5.45, P = 0.021) and hypertension (OR = 12.11, P = 0.007), but not with kidney size (P = 0.23). The APAT achieved good internal consistency (Cronbach's alpha coefficient = 0.91) and test-retest reliability (domain intra-class correlation coefficients ranging from 0.62 to 0.90). CONCLUSIONS: The APAT demonstrated good acceptability and reliability, and following further validation in a larger cohort could represent an invaluable tool for future ADPKD pain studies.Addenbrookes Charitable Trust Kidney Care UK British Renal Society Kidney Research U

    Recombination in Streptococcus pneumoniae Lineages Increase with Carriage Duration and Size of the Polysaccharide Capsule

    Streptococcus pneumoniae causes a high burden of invasive pneumococcal disease (IPD) globally, especially in children from resource-poor settings. Like many bacteria, the pneumococcus can import DNA from other strains or even species by transformation and homologous recombination, which has allowed the pneumococcus to evade clinical interventions such as antibiotics and pneumococcal conjugate vaccines (PCVs). Pneumococci are enclosed in a complex polysaccharide capsule that determines the serotype; the capsule varies in size and is associated with properties including carriage prevalence and virulence. We determined and quantified the association between capsule and recombination events using genomic data from a diverse collection of serotypes sampled in Malawi. We determined both the amount of variation introduced by recombination relative to mutation (the relative rate) and how many individual recombination events occur per isolate (the frequency). Using univariate analyses, we found an association between both recombination measures and multiple factors associated with the capsule, including duration and prevalence of carriage. Because many capsular factors are correlated, we used multivariate analysis to correct for collinearity. Capsule size and carriage duration remained positively associated with recombination, although with a reduced P value, and this effect may be mediated through some unassayed additional property associated with larger capsules. This work describes an important impact of serotype on recombination that has been previously overlooked. While the details of how this effect is achieved remain to be determined, it may have important consequences for the serotype-specific response to vaccines and other interventions

    Occupational Exposure to Endocrine-Disrupting Chemicals and Birth Weight and Length of Gestation: A European Meta-Analysis

    BACKGROUND: Women of reproductive age can be exposed to endocrine-disrupting chemicals (EDCs) at work and exposure to EDCs in pregnancy may affect fetal growth. OBJECTIVES: We assessed whether maternal occupational exposure to EDCs during pregnancy as classified by application of a job exposure matrix was associated with birth weight, term low birth weight (LBW), length of gestation, and preterm delivery. METHODS: Using individual participant data from 133,957 mother-child pairs in 13 European cohorts spanning births from 1994 to 2011, we linked maternal job titles with exposure to 10 EDC groups as assessed through a job exposure matrix. For each group, we combined the two levels of exposure categories (possible and probable) and compared birth outcomes with the unexposed group (exposure unlikely). We performed meta-analyses of cohort-specific estimates. RESULTS: Eleven percent of pregnant women were classified as exposed to EDCs at work during pregnancy based on job title. Classification of exposure to one or more EDC group was associated with an increased risk of term LBW (OR 1.25, 95%CI 1.04, 1.49), as were most specific EDC groups; this association was consistent across cohorts. Further, the risk increased with increasing number of EDC groups (OR 2.11 95%CI 1.10, 4.06 for exposure to 4 or more EDC groups). There were few associations (p < 0.05) with the other outcomes; women holding job titles classified as exposed to bisphenol A or brominated flame retardants were at higher risk for longer length of gestation. CONCLUSION: Results from our large population-based birth cohort design indicate that employment during pregnancy in occupations classified as possibly or probably exposed to EDCs was associated with an increased risk of term LBW

    Spontaneous DNA damage to the nuclear genome promotes senescence,redox imbalance and aging

    Accumulation of senescent cells over time contributes to aging and age-related diseases. However, what drives senescence in vivo is not clear. Here we used a genetic approach to determine if spontaneous nuclear DNA damage is sufficient to initiate senescence in mammals. Ercc1-/Δ mice with reduced expression of ERCC1-XPF endonuclease have impaired capacity to repair the nuclear genome. Ercc1-/Δ mice accumulated spontaneous, oxidative DNA damage more rapidly than wild-type (WT) mice. As a consequence, senescent cells accumulated more rapidly in Ercc1-/Δ mice compared to repair-competent animals. However, the levels of DNA damage and senescent cells in Ercc1-/Δ mice never exceeded that observed in old WT mice. Surprisingly, levels of reactive oxygen species (ROS) were increased in tissues of Ercc1-/Δ mice to an extent identical to naturally-aged WT mice. Increased enzymatic production of ROS and decreased antioxidants contributed to the elevation in oxidative stress in both Ercc1-/Δ and aged WT mice. Chronic treatment of Ercc1-/Δ mice with the mitochondrial-targeted radical scavenger XJB-5–131 attenuated oxidative DNA damage, senescence and age-related pathology. Our findings indicate that nuclear genotoxic stress arises, at least in part, due to mitochondrial-derived ROS, and this spontaneous DNA damage is sufficient to drive increased levels of ROS, cellular senescence, and the consequent age-related physiological decline
